How do you create 2D animation (arrows, text, etc)?

For a presentation, I'd like to animate some simple 2D vectors over an image (An arrow pointing at something, highlighting areas, etc). What is the best way to do this? Do you just use flash?

 

Also, for future reference, what software do you use to overlay 2D animation on a video/walkthrough? Will Premiere do that or are there better solutions?

I think After Effects is what you are looking for. It can animate splines and text ect... It is like having Photoshop for Videos that can also animate with key frames. That's probably selling it way short, but it's a great tool. In the end you just compress whatever it is into a video file. At our studio we take the compressed footage into Premier to edit to music, but that really just because we separate shots so that a team can work on them. AE can do this too it's just more cumbersome.
